While a reboot is not necessarily of a particular value other than what Jan-Erik mentioned, I would use that time to create an image copy of the disks.  This is the safest way to create a defragmented disk.  Unless you are running DFG, the drives will become fragmented over time.  While in most instances this will not present a problem, directory expansions may be impacted with little contiguous space.
