No, the cluster size should not be an issue.  The error is with the tape 
drive, not the disk.

As Steven asked, what TYPE of tape drive?

Cleaning the tape drive would be something to try.

Trying another tape drive.

Do you have other backups?  On tape?  On disk?

You have just stated a good reason to use RAID 1, mirrored disks.
